The race starts on Main Street. From there runners bear right along cobbles and through Sun Street towards Penistone Hill. After a brief uphill section, they pass the cricket ground and along Lee Lane to the nearby reservoir. The next four miles is on tarmac, over a stream and across an open moor to Withens Pub. The route continues across countryside, passing near Gatehouse and via Houghgate and Brookhouse reaching the halfway mark at the junction with the A629.

The inward half starts along a footpath to Denholme Beck. Under a railway bridge to a reservoir, through allotments and over a viaduct, the route carries on through forest and across moorland to Hainworth Village. From there, runners pass along Black Moor Road, the nearby farmhouse and onto Penistone Hill. Then the last 1.2 miles onto the finish line.
